Hidralazinin SH-SY5Y hücre hattında PTZ ile indüklenen nörotoksisite üzerindeki nöroprotektif etkisinin araştırılması

ÖZET

Amaç: Akut nöbet, bir grup kortikal nöronun aşırı ve senkronize deşarjlarına bağlı olarak ortaya çıkan ve klinikte gelip geçici olmakla birlikte hipokampusta nöron kaybına neden olan önemli bir nörolojik rahatsızlıktır. Oksidatif stres, nöbet sonrası meydana gelen hipokampal hasarlanmada etkin role sahiptir. Bu çalışmanın amacı, SH-SY5Y hücre hattında pentilentetrazol (PTZ) ile oluşturulan akut nöbet modeline hidralazin uygulayarak sinir koruyucu etkilerini araştırmaktır.

Yöntem: SH-SY5Y hücre kültüründe kontrol grubu, PTZ grubu, hidralazin grubu ve hidralazin+PTZ grubu olmak üzere 4 grup planlanmıştır. Kontrol grubuna herhangi bir uygulama yapılmazken hidralazin+PTZ grubuna çeşitli konsantrasyonlarda (6.25; 12.5; 25; 50 ve 100 µg/ml) hidralazin uygulanıp 24 saat sonra PTZ uygulanmıştır. XTT hücre canlılığı testi ile ilacımızın toksisite üzerine etkisi belirlenmiştir. Hücre süpernatantlarındaki doku TOS konsantrasyonları otomatik test yöntemi ile ölçülmüştür. Analizin verimliliği için hidrojen peroksit kullanılmıştır.

Bulgular: XTT hücre canlılığı sonuçları değerlendirildiğinde, hidralazin, toksisite oluşturulan SH-SY5Y hücrelerinde sinirsel (nöronal) canlılığı etkilememiştir. Ek olarak, hidralazin TOS seviyeleri üzerinde de kayda değer bir etki yapmamıştır (P>0.05).

Sonuçlar: Elde edilen veriler, çalıştığımız dozlardaki hidralazinin, PTZ ile indüklenen SH-SY5Y hücre hattı nörotoksisitesinde sinir koruyucu etkiye sahip olmadığını göstermiştir.

ABSTRACT

Objective: Acute seizure is an important neurological disorder that occurs due to excessive and synchronized discharges of a group of cortical neurons and causes neuronal loss in the hippocampus, although it is clinically temporary. Oxidative stress has an effective role in hippocampal damage that occurs after seizures. The aim of this study is to investigate the neuroprotective effects of hydralazine in an acute seizure model created with pentylenetetrazole (PTZ) in the SH-SY5Y cell line.

Method: In the SH-SY5Y cell culture, 4 groups were planned as control group, PTZ group, hydralazine group and hydralazine + PTZ group. While no application was made to the control group, hydralazine was applied to the hydralazine + PTZ group at various concentrations (6.25; 12.5; 25; 50 and 100 µg / ml) and PTZ was applied 24 hours later. The effect of our drug on toxicity was determined by XTT cell viability test. Tissue TOS concentrations in cell supernatants were measured by the automated test method. Hydrogen peroxide was used for the efficiency of the analysis.

Results: When evaluating XTT cell viability results, hydralazine did not affect neuronal viability in toxicity induced SH-SY5Y cells. In addition, hydralazine did not have a significant effect on TOS levels (P>0.05).

Conclusions: The data obtained showed that hydralazine at the doses we studied did not have a neuroprotective effect on the SH-SY5Y cell line neurotoxicity induced by PTZ.
